The mutant Asp38Arg of the lightdriven protonpump bacteriorhodopsin (BR) was investigated with a combination of UV-VIS, FTIR and X-ray experiments. According to measurements at different pHs - with and without GuaHCI - it could be shown that also bythis mutant the big changes in the tertiärstructure take place at the M1 to M$_{2}$ transition. On the basis of the pH dependent accumulation of the different M-species it can in addition be concluded that the big changes in the tertiärstructure could only take place after a redistribution of charges in the BR-molecule. Dynamic light scattering was used to investigate the properties of BR-detergentmicelles to get an improvement of the BR crystallisation conditions. Three detergent mixtures could be found witch should lead to an improvement in the crystallisation of BR because of smaller BR-detergentmicelles and an increased stability of BR in this micelles. The three detergent mixtures and also their components were characterised according to changes in the concentration of the detergent and the ion strenght of the solution. |
