Polyclonal and monoclonal antibodies against photosensory membrane preparations of the crayfish (Astacus leptodactylus) were obtained and are described. Their specific interaction can be used in functional studies of the Astacus and Limulus photoreceptor cello Preparations of Astacus photosensory membrane yielo about 5-8 $\mu$g protein per retina. As characterized by SDS-gel electrophoresis, the predominant protein is the visual pigment (40 - 50 % of the total protein content). In addition, there are about 9 other major protein bands as demonstrated by Coomassie Brilliant Blue staining, as weil as a number of minor protein bands which can be detected by silver stain. A solid phase, indirect radio immune assay employing polyvinylchloride microtiter plates was set up as a rapid and sensitive test for antibodies. Since no purified antigens were available, a sonicated preparation of whole illuminated rhabdoms, containing not only photosensory membrane but also cytoplasmic proteins of the rhabdom, was used for this test. |
